Understanding NK cell biology for harnessing NK cell therapies: targeting cancer and beyond

چکیده

Gene-engineered immune cell therapies have partially transformed cancer treatment, as exemplified by the use of chimeric antigen receptor (CAR)-T cells in certain hematologic malignancies. However, there are several limitations that need to be addressed target more types. Natural killer (NK) a type innate represent unique biology surveillance. In particular, NK obtained from heathy donors can serve source for genetically engineered therapies. Therefore, NK-based therapies, including cells, CAR-NK and antibodies induce antibody-dependent cellular cytotoxicity emerged. With recent advances genetic engineering techniques, cell-based become promising approaches wide range cancers, viral infections, senescence. This review provides brief overview characteristics summarizes diseases could benefit addition, we discuss preclinical clinical investigations on adoptive transfer agents modulate activity.

Investigation of NK Cell Population in Peripheral Blood and Tumor Lesions of Patients with Breast Cancer

Background: Alteration in peripheral blood lymphocytes (PBLs) is usually investigated to provide an evidence of the host immune responses to tumor antigens. The tumor infiltrating NK cells interact most closely with the tumor cells and more accurately reflect tumor host interactions. Objective: To analyze peripheral blood and tumor associated Natural Killer (NK) cells in patients with breast ca...

NK Cell and CD4+FoxP3+ Regulatory T Cell Based Therapies for Hematopoietic Stem Cell Engraftment

Allogeneic hematopoietic cell transplantation (HCT) is a powerful therapy to treat multiple hematological diseases. The intensive conditioning regimens used to allow for donor hematopoietic stem cell (HSC) engraftment are often associated with severe toxicity, delayed immune reconstitution, life-threatening infections, and thus higher relapse rates. Additionally, due to the high incidence of gr...

Prognostic factors for mature natural killer (NK) cell neoplasms: aggressive NK cell leukemia and extranodal NK cell lymphoma, nasal type.

BACKGROUND Patients with natural killer (NK) cell neoplasms, aggressive NK cell leukemia (ANKL) and extranodal NK cell lymphoma, nasal type (ENKL), have poor outcome. Both diseases show a spectrum and the boundary of them remains unclear. The purpose of this study is to draw a prognostic model of total NK cell neoplasms. PATIENTS AND METHODS We retrospectively analyzed 172 patients (22 with A...

Harnessing Soluble NK Cell Killer Receptors for the Generation of Novel Cancer Immune Therapy

The natural cytotoxic receptors (NCRs) are a unique set of activating proteins expressed mainly on the surface of natural killer (NK) cells. The NCRs, which include three members; NKp46, NKp44 and NKp30, are critically involved in NK cytotoxicity against different targets, including a wide range of tumor cells derived from various origins. Even though the tumor ligands of the NCRs have not been...
